Her main skills include exceptional agility and acrobatic abilities, being a former athlete and gymnast. She is also a capable hand-to-hand combatant, employing an anarchic and unpredictable style that often catches her opponents off guard.Harley is skilled in using various weapons, with her signature large wooden mallet being the most recognizable. She often also uses pistols, explosives, and other improvised weapons, which she wields in an imaginative and destructive manner. Her intelligence as a former psychiatrist also allows her to have a deep understanding of human psychology, which she uses to manipulate and confuse her opponents.

Harley Quinn has become a pivotal and immensely popular member of the Suicide Squad, bringing a dynamic sense of chaos and unpredictability to the team. Despite her initial devotion to The Joker, Harley has evolved and distanced herself from him, developing her own relationships and motivations. Within the Suicide Squad, she often forms unexpected alliances and friendships, with her most notable dynamic being with Deadshot, who often acts as a strange mentor or friend to her. Her relationship with Poison Ivy is also iconic, evolving into a deep romantic connection, offering Harley a healthier and more supportive relationship. Harley brings an element of madness and intelligence to the team, often using her superficial insanity to be underestimated by opponents.

Although The Joker is Harley Quinn's most significant and defining adversary (even though their relationship is complex and often toxic), she has faced other notable enemies. The Batman Family as a whole, especially Batman and Robin, are frequently her opponents, as they try to stop her criminal endeavors. Amanda Waller can also be considered an adversary, as her manipulative tactics and her intent to use Harley as an expendable member of Task Force X create a constant conflict. Furthermore, she has clashed with other Gotham City villains who compete for territory or influence, or even with former "friends" who turn against her. As Harley has developed her own autonomous path, she has also gained new adversaries related to her more anti-heroic attempts to do good, or at least be less villainous.

Harley Quinn has become incredibly popular in non-comic adaptations, particularly in film and television. The most iconic portrayal is by Margot Robbie in the DCEU films. She debuted in "Suicide Squad" (2016), where she stood out despite the film's mixed reviews. She reprised the role in the critically successful "Birds of Prey" (2020), which focused on her liberation from The Joker and the formation of her own team. She also appeared in James Gunn's "The Suicide Squad" (2021), which further established her as an independent, chaotic, and sympathetic anti-hero. Kaley Cuoco voices Harley Quinn in the successful HBO Max animated series "Harley Quinn" (2019-present), which has received widespread acclaim for its dark humor, subversive tone, and the development of her relationship with Poison Ivy.
