The basic premise of Hell's Paradise is set in Edo period Japan and follows Gabimaru the Hollow, a legendary ninja assassin renowned for his unparalleled ruthlessness and inability to die, even though he desires death. Sentenced to execution, Gabimaru refuses to die, as he still has a hidden desire to reunite with his wife. He is given a last chance by Sagiri Yamada Asaemon, an executioner from the renowned Asaemon family of executioners. Gabimaru, along with other dangerous convicts, is sent to a mysterious and deadly island, known as Shinsenkyo or "Paradise," with the mission to find the Elixir of Immortality. The competition among the convicts, the island's deadly traps, and the bizarre, supernatural creatures inhabiting it make the mission exceedingly difficult.

Gabimaru is accompanied on his dangerous island mission by various key characters, both convicts and executioners. Sagiri Yamada Asaemon is the female executioner tasked with overseeing Gabimaru. Although initially uncertain of her executioner abilities, Sagiri is a skilled swordswoman and strives to understand Gabimaru's humanity. Other important convicts include Aza Chobei, a ruthless bandit with astonishing survival skills and his brother. Yuzuriha, a clever and manipulative kunoichi who uses her charm and poisons. And Fuchi Yamada Asaemon, a competent executioner and member of the Asaemon family, who supervises another convict. These characters, with their diverse motivations and abilities, create a dynamic ensemble that enhances the plot's intensity and complexity.

Hell's Paradise explores a multitude of dark and existential themes. Central to it is the search for the value of life and the concept of "immortality," not just biologically, but also through achieving goals and connecting with others. The theme of human nature is dominant, exploring the limits of cruelty, survival, and the potential for change and redemption. The series addresses the choice between death and life, and the motivation that love can provide even to the most corrupt.
