The basic premise of Assassination Classroom is highly unique and original. The story begins with a powerful, octopus-like alien creature, capable of moving at Mach 20 speed, which destroys 70% of the moon. This being, named Koro-sensei (a combination of "Koro," meaning "unkillable," and "sensei," meaning "teacher"), announces that he will destroy Earth in one year, but first requests to become the teacher of Class 3-E at Kunugigaoka Junior High School. The government agrees, offering a huge bounty to any student who can assassinate him before the end of the school year. Thus, the students of the "failing" class, considered "failures" by the school, take on the role of would-be assassins, while Koro-sensei teaches them not only academics, but also how to become better people and assassins, fostering a peculiar and heartwarming teacher-student relationship.

Class 3-E, although considered the "E-class failures," consists of many unique and developing characters, each with their own skills and issues. Nagisa Shiota is the seemingly quiet and observant student with an unpredictable assassination talent and a hidden analytical skill. Karma Akabane is the most talented and intelligent student in the class, but also extremely unruly and sadistic, who enjoys testing Koro-sensei. Kaede Kayano is a cheerful and supportive student, who develops a deep connection with Koro-sensei and hides her own secrets. Tadaomi Karasuma is the government agent overseeing the class and teaching them martial arts. Irina Jelavic is a professional assassin hired to kill Koro-sensei and teaches English. These characters, along with the rest of the students, develop deep friendships and learn to cooperate towards their common goal, while also maturing as individuals.

Assassination Classroom explores a multitude of themes that combine comedy, action, and drama, with a profound message about education. Central to it is the concept of "education" and how a true teacher can inspire their students, even if the ultimate goal is their assassination. The theme of self-esteem and overcoming rejection is dominant, as the students of Class 3-E, considered "outcasts," gain confidence and discover their potential. The unique teacher-student relationship, friendship, cooperation, and the search for personal purpose are also explored. The series addresses the concept of "assassination" in an unexpected way, as students develop skills, but simultaneously learn about life and human values.
